Serum from Apo E−/− mice exposed to MVE alters BBB transport activity in mouse BBB co-culture. BBB function, as shown by P-glycoprotein activity, was assessed by measuring the passage of Vinblastine (0.1 μM) across BBB mouse co-cultures. Serum (1/30 dilution in media) from Apo E−/− mice exposed to either filtered air (FA) or mixed vehicular exhaust (MVE, 100 PM μg/m3), for 30 days, was applied to the apical endothelial layer transwell for (A) 4 hrs or (B) 24 hrs. [3H]-Vinblastine was then measured in both endothelial and glial well supernatants by scintillation counting at 1 hr (37°C) and resulting clearance was calculated. N = 3 per group, run in duplicate. *p < 0.050 compared to FA control.
