Effects of multiple different pulmonary exposures on lung mRNA levels of Irf7 and associated pattern recognition receptors. Lung RNA extracted from previously published exposures was assessed post-hoc with respect to the new findings from this study. Male C57BL/6 J mice were exposed to 340 μg of gas metal arc – stainless steel (GMA-SS), 340 μg gas metal arc – mild steel (GMA-MS), 40 μg of multi-walled carbon nanotubes (MWCNT), or 40 μg single-walled carbon nanotubes (SWCNT) (n = 5 or 6 per group for all exposures) by pharyngeal aspiration and lungs were harvested 24 hr post-exposure [15, 19]. Relative mRNA levels of Irf7 (interferon regulatory factor 7) and the pattern recognition receptors Ddx58 (DEAD box polypeptide 58; RIG-I) and Ifih1 (interferon induced with helicase C domain; MDA5) were increased in the lungs of GMA-SS exposed mice only in reference to sham (dotted line). *p<0.05 vs sham.
